A-YPS Test-Kit for the detection of progestogenic substances

In our uniquely developed A-YPS (Arxula Yeast Progesterone Screen) test method the best properties of all effect-based and not ELISA-related methods are combined.  Hence, this test kit enables you to determine the progestogenic activity of aqueous samples already at a concentration of estrogens and estrogenic substances  in a single-digit ng/L range.

The robust measurement method is insusceptible to microbial flora and therefore can be applied to complex samples without significant pretreatment (for example, samples from sewage treatment plants or those with high salt content). Sterile working conditions are not required. Using standardized production conditions, we guarantee the consistent quality our test organism. The ready-to-use yeast offers the benefit of significantly reducing the effective workload of your laboratory. The recombinant test organism, Arxula adeninivorans, can handle samples that have different salinity and are spiked with progestogenic substances on a single plate within approximately 26 hours.

The A-YPS test is based on a genetically modified strain of the A. adeninivorans yeast which has been modified to bind with the human progesterone receptor, hPR-B. Binding of progestogenic substances to the receptor subsequently activates production of a secretory reporter enzyme. The level of enzyme activity correlates to the total concentration of progestogenic substances in the sample. With the addition of the chromogenic substrate p-nitrophenyl phosphate, enzyme activity can be measured photometrically. Progesterone is used as a reference standard for calibration and quantification.

The detection is carried out photometrically after an enzymatic reaction with a chromogenic substrate (p-nitrophenylphosphate).
