This cell-based screening method determines the activity of single substances and the cumulative activities of a substance mix which are mediated by Arylhydrocarbon (Ah) receptors. It is intended to be used for testing of substances like meat and fish. Sterile conditions are not required for this test. The ready-to-use yeast offers the benefit of significantly reducing your workload and workflow. By using the recombinant test organism Arxula adeninivorans you can test your samples within approximately 26 hours for the presence of Ah-receptor affine ligands. Using standardized production conditions, we ensure the consistent quality our test organism.

The A-YDS test is based on a genetically modified strain of the A. adeninivorans yeast in which genome the gene for the human Arylhydrocarbon receptor (hAhR) as well as the gene for the human Arylhydrocarbon receptor nuclear trankslocator (hARNT) was integrated. If a substance with chemical affinity (ligand) is present, the reporter enzym (phytase) is produced throught the interaction of the ligand the receptor dimer hAH and hARNT. The amount of the reporter enzyme correlates to the total concentration of substances in the sample which interact with the receptors. With the addition of the chromogenic substrate p-nitrophenyl phosphate, the concentration of the reporter enzyme can now be measured. β-Naphthoflavone is used as a reference standard for calibration and quantification.
